Overview of PPI Dendrimers

There are many types of dendrimers, among which polyamido-amines (PAMAM), poly-L-lysine (PLL) and poly(propyleni-mine) (PPI) are currently commercialized and widely used in biomedicine. Among them, PPI dendrimers are promising three-dimensional polymers for pharmaceutical and biomedical applications, generally characterized by the presence of primary amine terminal groups and tertiary propylene amines inside the PPI structure. The main mechanism of action of PPI dendrimers is to increase the solubility of conjugated drugs through electrostatic interactions, and their main advantages include easy surface modification, allowing for high-substituted dendrimers, and drug delivery versatility and high functionality.

Applications of PPI Dendrimers

Dendrimers are highly branched, fully monodisperse macromolecules with a precisely controlled chemical structure. The term “dendrimers” is a combination of two Greek words "dendron" and "meros", which translate to tree and parts. The first paper on dendrites was published in 1978 by Vögtle and coworkers, who created dendritic structures by using divergent synthesis. The convergent synthesis method that followed was proposed by Hawker and Frechet in 1990. As a synthetic polymer, dendrimers emerge from a focal point with repeated branching units that expose many anionic, neutral, or cationic terminal functions on their surface, resulting in dendrimers that become hydrophobic or hydrophilic compounds.
