Prokaryotic expression system

Advantages Clear genetic background multiplication, low cost, high expression 2H/13C/15N isotope labeling technology is stable and widely applicable.
Limitations
  • No transcriptional and post-translational processing functions, difficult to form correct spatial conformational folds.
  • Expressed proteins are often insoluble and form inclusion bodies.
  • Applicable to simple eukaryotic proteins and isotope-labeled proteins.

Yeast expression system

Advantages
  • The expression process does not produce toxins.
  • Processing of expression products can be carried out to maintain the activity and stability of the products.
  • Expression product is secreted to extracellular, which is conducive to purification.
Limitations
  • Exogenously expressed proteins are easily degraded by their own secreted proteol.
  • The fermentation cycle is relatively long and easy to be contaminated.
  • Suitable for expressing secreted proteins and proteins that are cytoplasmically localized and not modified by glycosylation.

Mammalian expression system

Advantages Expression of glycosylated proteins and normal reconstitution of multiple subunits. High density and large culture volume.
Limitations A long time is required to obtain highly expressed cell lines and high cost for large-scale culture.
  • Suitable for expression of secretory proteins, enzymes, transmembrane proteins ion channels, etc. The protein structure is close to the natural state.

Insect expression system

Advantages
  • Does not cause harm to humans or environmental pollution.
  • Short vector construction period.
  • Ideal for the expression of large segments of DNA.
  • Can express multiple genes simultaneously in the same cell.
Limitations Recombinant proteins are easily degraded.
  • Suitable for expression of secreted proteins and polyprotein mixtures.
