Introduction This gene encodes a protein belonging to the raf/mil family of serine/threonine protein kinases. This protein plays a role in regulating the MAP kinase/ERKs signaling pathway, which affects cell division, differentiation, and secretion. Mutations in this gene are associated with cardiofaciocutaneous syndrome, a disease characterized by heart defects, mental retardation and a distinctive facial appearance. Mutations in this gene have also been associated with various cancers, including non-Hodgkin lymphoma, colorectal cancer, malignant melanoma, thyroid carcinoma, non-small cell lung carcinoma, and adenocarcinoma of lung. Revival Cells are cultured as a monolayer at 37°C in a humidified atmosphere with 5% CO2. Cells should be passaged every 3-5 days. Split at 80-90% confluency, approximately 1:6-1:10.
Culture Medium DMEM/F-12 including 2.5 mM L-glutamine and 15 mM HEPES, supplemented with 5% horse serum, 10 µg/mL insulin, 20 ng/mL hEGF, 0.5 µg/mL hydrocortisone, 0.1 µg/mL cholera toxin.
