Description
Liposomes are spherical self-closing structures formed by one or more concentric lipid bilayers with an encapsulated aqueous phase in between, and between the bilayers are composed of natural or synthetic lipids. Due to their size, hydrophobic/hydrophilic properties, and biocompatibility, liposomes are potential drug delivery systems. However, the low solubility and short in vivo half-life of liposomes limit their application in pharmaceutical research. To solve these problems, PEGylation of liposomes is a good solution. After PEGylation, the hydrophilicity of the liposome surface increases, and the affinity with phagocytes decreases, so it can escape the recognition of the reticuloendothelial system and reduce the capture of liposomes. PEGylation can reduce the occurrence of immunogenicity and antigenicity of liposomes. In addition, PEG liposome encapsulation also affects the physicochemical properties of active molecules. It can reduce the liver clearance rate and prolong the half-life of the drug, thereby reducing the dosage and frequency of medication in the experiment; at the same time, it can reduce the adverse reactions of the drug while ensuring the efficacy of the drug.
