Description

Agarose bound soy lectins are prepared using affinity purified lectins. Soy lectins are a closely related family of isolates consisting of four subunits of roughly equal size. The molecular weight of the glycoprotein is about 120kda, and the isoelectric point is near pH 6.0. SBA preferentially binds to oligosaccharide structures of n-acetylgalactosamine with alpha - or beta - junctions at the end, and to a lesser extent to galactose residues. Binding can be prevented by substitution on the second-to-last sugar, such as the focal point attached to the second-to-last galactose in the type B blood substance.

Recommended dilution
Wash gel thoroughly with buffer before use to remove sugar added to stabilize the lectin. Recommended product for eluting glycoconjugates bound to this agarose-lectin: Glycoprotein Eluting Solution, Cat. No. ES-2100. Alternatively, 0.2 M N-acetylgalactoseamine in buffered saline can be used. After use, wash the gel with several column volumes of buffered saline, then resuspend gel in buffered saline containing 0.08% sodium azide for storage.
