Plant-specific proteins include functional proteins, enzymes, post-translational modifications (PTM), and low-specificity target proteins. Accurate and sensitive quantification of specific proteins is the key to most protein assays. Our methods for protein-specific quantification include enzyme-linked immunosorbent assay (ELISA), western blot, and mass spectrometry.

Quantification of plant-specific protein based on mass spectrometry

We offer mass spectrometry-based quantification of plant-specific proteins, including MALDI-TOF and ESI-MS.

Method Description Sample Requirements
MALDI-TOF MALDI is suitable for the quantification of proteinaceous samples for which ionization produces fragmentation. Protein purity > 90%; protein content > 5μg, concentration > 0.1μg/μl.
Bidirectional gel electrophoresis spots of staining or silver staining are visible.
The SDS-PAGE strip is a single protein, and the staining or silver-staining bands are visible.
ESI-MS ESI-MS can analyze protein samples that are strongly polar, difficult to volatilize, or thermally unstable. Protein content > 5μg, concentration > 0.1μg/μl. Buffer does not contain detergent NP40, Triton X-100, etc.
Cow staining or silver staining bands of SDS-PAGE adhesive strips are visible.

We provide enzyme-linked immunosorbent assays (ELISA) that can easily quantify a large number of samples for qualitative and quantitative analysis.

Method Description Sample Requirements
Direct ELISA Direct ELISA requires the preparation of various coupled antibodies and is suitable for the quantification of antigens. Plant tissue sample > 10g.
Antigen or antibody concentration > 1-20μg/ml, volume > 200μl.
Indirect ELISA Indirect ELISA is commonly used to quantify total antibodies.
Sandwich ELISA Sandwich Elisa is suitable for the quantification of antigens or antibodies in complex samples.

Quantification of plant-specific protein based on nanoparticles and nanopores

Method Description Sample Requirements
Nanoparticle Sensors Nanoparticles are chemically cross-linked to proteins, allowing the detection and quantification of specific proteins, such as antibodies, peptides, and ligands, based on absorption spectra. Plant tissue sample > 5g.
Protein concentration > 1-20μg/ml,  volume > 200μl.

Method Description Sample Type
Western Blot Western blotting can only be used for semi-quantitative or relative quantitative analysis. SDS-PAGE protein bands, 2D protein spots, pull-down, and Co-IP samples.
