The kit provides rapid separation of fresh/frozen plant soft tissues, especially leaves.
Using only 100-200 mg of starting material, the kit uses a dedicated filter box plus our proprietary buffering system to obtain four subcellular components (cytoplasm, nucleus, chloroplast and organelles) in about 1 hour. Subcellular components can be used for many applications, including but not limited to protein transport, nucleoprotein extraction, and nucleic acid purification/sequencing. Highly enriched nuclei can be used for proteomics, flow cytometric analysis, DNA sequencing, and protein-protein/protein-DNA interaction studies. The subcellular separation protocol involves grinding the tissue to release the plant cells and then destroying the cell membrane by rapidly passing through the filter. The different subcellular components are obtained by differential centrifugation using only a table centrifuge. The kit has been tested for many species such as Arabidopsis, spinach, peas and canola.
