Description

PA200(proteasome activator PA200) is a related component of the proteasome that specifically recognizes acetylated histones and promotes ATP-independent and ubiquitin degradation of core histones in DNA damage responses. It also recognizes and binds acetylated histones through its bromine domain-like (BRDL) region and activates the proteasome by opening a gated channel into which the substrate enters. Through its C-terminal binding to the core proteasome, C-terminal and proteasome ATPase occupy the same binding site, opening the closed structure of the proteasome through the activity gating mechanism. Participates in DNA damage reactions: binds to acetylated histone proteins and promotes histone degradation. MG132 is a specific, potent, reversible and cell-permeable proteasome inhibitor, and the protein levels increase after exposure to MG132.
