Cell-Penetrating Peptides and Pep-1

Cell-penetrating peptides (CPPCs) are a class of short peptides that can carry a wide range of biomolecules of different sizes and properties (including small molecules, dyes, peptides, proteins, and nucleic acids) into cells without relying on classical cytokinesis for their penetration ability. Transactivating transcriptional activator (TAT), from human immunodeficiency virus 1 (HIV-1) was the first peptide identified to enter cells in a non-toxic and efficient manner. CPPCs can act as molecular delivery vehicles to deliver different types of cargo molecules across the plasma membrane. Their applications in biomedicine include the delivery of drugs for the treatment of different diseases (cancer and viral inhibitors), as contrast agents for cellular markers.

Pep-1 is an amphipathic cell-penetrating peptide consisting of a tryptophan-rich hydrophobic structure and a lysine-rich hydrophilic structural domain. Pep-1 rapidly localizes to the nucleus by endocytosis-independent internalization and efficiently delivers various peptides and proteins in a fully bioactive form in cells.

Dendrimer-Pep-1 Conjugation

Interleukin-13 (IL-13) plays an important role in regulating the immune response and immune microenvironment not only in normal physiological situations but also in cancer. Interleukin-13 receptor α2 (IL-13Rα2), one of the subunits of the interleukin-13 receptor, is overexpressed in glioma cells and is an important target for glioma therapy. Pep-1 is a linear peptide with 9 amino acid residues (CGEMGWVRC), a specific ligand for IL-13Rα2, and has a high affinity to IL-13Rα2. Dendrimers are widely used in biomedicine for their hyperbranch, monodispersion, threedimension, well-defined molecular weight and host-guest entrapment properties. Dendrimers are often used as carriers to deliver drugs. When combined with Pep-1, a cell-penetrating peptide, dendrimers can target glioma cells through IL-13Rα2-mediated endocytosis and cross various physiological barriers such as the blood-brain barrier (BBB) and the blood-brain tumor barrier (BBTB) to achieve cancer therapy.
