Structure of Tuftsin

Immunomodulation includes the stimulation and suppression of the immune system by various substances called immunomodulators, designed to modulate the body's immune response to different etiologies, and is an important therapeutic strategy. As an immunomodulator, tuftsin is a tetrapeptide (Thr-Lys-Pro-Arg, TKPR) located in the Fc structural domain of the immunoglobulin G heavy chain (residues 289-292). Tuftsin is involved in a variety of biological functions related to the immune system and has broad-spectrum immune activities such as migration, phagocytosis, bactericidal, and tumor suppression. Due to structural similarities, tuftsin binds specifically to macrophages, monocytes and polymorphonuclear leukocytes. In addition, tuftsin alleviates symptoms, raises blood pressure, and inhibits lymphatic vasoconstriction.

Dendrimer-Tuftsin Conjugation for Targeted Treatment of Cancer

Dendrimers are a class of polymers with nanoscale structures that are mainly used in biomedical applications. There are several types of dendrimers, among which poly(propyleni-mine) (PPI) dendrimers are promising three-dimensional polymers for pharmaceutical and biomedical applications, generally characterized by the presence of primary amine terminal groups and tertiary propylene amines inside the PPI structure. The main mechanism of action of PPI dendrimers is to increase the solubility of conjugated drugs through electrostatic interactions, and their main advantages include easy surface modification, allowing for high-substituted dendrimers, and drug delivery versatility and high functionality. When PPI dendrimers are bound to tuftsin, tuftsin can specifically bind to macrophages, monocytes and polymorphonuclear leukocytes due to structural similarities. In addition, tuftsin shields the cationic moiety of the dendrimers and reduces cytotoxicity. Therefore, when dendrimers are conjugated with tuftsin, they play an important role in HIV-targeted therapy, improving the efficacy of tumor chemotherapy and reducing systemic drug toxicity.
