CD BioSciences has advanced equipment and experienced scientists to provide you with high-quality dendrimer PEGylation service.
Introduction into PEGylation
Polyethylene glycol (PEG), also known as polyethylene oxide (PEO) or polyoxyethylene (POE), is an oligomer or polymer of ethylene oxide. The term PEGylation was first coined by Davis and Abucvhowsky in 1970 and is the joining of two molecules using bioconjugation, one of which is a biomolecule and the other is a PEG. When PEGylation of biomolecules occurs, it has the following advantages.
- Bispecific interactions between proteins or nanomaterials and cells or tissues can be effectively reduced or prevented.
- When drug carriers, such as dendrimer, undergo PEGylation, they can improve biocompatibility and stability, increase drug circulation time, reduce toxicity, and decrease drug side effects.
Dendrimer PEGylation
Dendrimer PEGylation refers to the attachment of PEG to the surface of dendrimers by different means. When dendrimer PEGylation occurs, it not only reduces toxicity, but also increases biocompatibility and in vivo circulation time.
Methods for Dendrimer PEGylation
There are 3 different methods to achieve dendrimer PEGylation.
- Direct coupling of PEG to the surface of the dendrimer.
- Attaching the PEG as a linker to the surface of the dendrimer to use it as a drug delivery vehicle after being attached to the drug.
- Using cross-linking agents to PEG the surface of the dendrimer to reduce the spatial resistance.
Advantages of Dendrimer PEGylation
When the dendrimer undergoes PEGylation it has the following advantages.
- Reduce Cytotoxicity: Dendrimer PEGylation can shield the positive charge of surface functional groups and inhibit the electrostatic interaction between cell membrane and surface cations of dendrimer.
- Improved Pharmacokinetics: When the dendrimer is polyethylene glycolized as a drug carrier, it can not only increase the loading capacity of the drug, but also improve the biodistribution of the drug, increase the circulation time and reduce the side effects of the drug.
- Other Advantages: Some dendrimers have non-immunogenicity, non-antigenicity, targeting and biocompatibility after PEGylation.
