Dendrimer-Nanoparticle Conjugation for Gene Therapy
Nanoparticles, including carbon nanotubes, graphene, quantum dots, gold nanoparticles, magnetic nanoparticles, upconversion nanoparticles, and silicon nanomaterials, are widely used for diagnosis and treatment of many diseases. Different nanomaterials have different unique properties, such as carbon nanotubes, graphene, and gold nanorods with photothermal activity, Fe3O4 nanoparticles with magnetic properties, and quantum dots and upconversion nanoparticles with fluorescent properties. Dendrimer can be easily conjugated with these nanoparticles to prepare less cytotoxic multifunctional materials for gene delivery. In future studies, nanoparticle-modified dendrimers play an important role in combination therapy (gene therapy, chemotherapy, photothermal therapy, etc.) for cancer treatment.

- PAMAM Dendrimer-Nanoparticle Conjugation
Dendrimers conjugated with nanoparticles have promising applications in gene delivery. For example, PAMAM dendrimers functionalized by multiwalled carbon nanotubes or carbon nanohorns can be used for efficient delivery of DNA, siRNA, and antisense oligonucleotides (asODN). Gold nanorods have an absorption peak around 800 nm, which can be used to monitor their location in cancer cells and improve the efficacy of gene transfection by near-infrared light irradiation. Modification of PAMAM dendrimers with gold nanorods enables delivery of short hairpin RNA (shRNA) into MCF-7 cells to kill cancer cells through a combination of gene therapy and photothermal ablation. In addition, PAMAM-dendrimer-encapsulated gold nanoparticles not only improved the transfection efficiency to several cell lines but also reduced the toxicity to cells. - PPI Dendrimer-Nanoparticle Conjugation
When PPI is conjugated with gold nanoparticles, the gold nanoparticles help the dendrimer to encapsulate the siRNA into nanoparticles but are not included in the yielding polyplexes. The complex can effectively deliver siRNA to cancer cells and silence their target mRNAs. In addition, the silencing effect of low generation dendrimer stabilized gold nanoparticles on mRNA is superior to that of high generation dendrimer.
