Dendrimers and Gene Therapy

Gene therapy is a promising option for the treatment of diseases such as genetic disorders, viral infections, and cancer. The main obstacle to gene therapy at present is the lack of safe and efficient gene vectors. Gene vectors are divided into viral and non-viral vectors. Viral vectors have great efficacy for both in vivo and in vitro gene delivery, but suffer from difficulties in mass production, high production costs and safety issues. Therefore, various non-viral vectors (including polymers, liposomes, exosomes, and nanomaterials) have been developed for gene delivery. Among the non-viral vectors, polymers are more attractive due to their structural, compositional and property diversity. When used as gene carriers, they offer advantages such as lack of immunogenicity, facile of synthesis, degradability, and ease of modification by functional ligands. Among the cationic polymers, dendrimers are unique nanoscale synthetic macromolecules with highly branched and spherical structures. When used as gene delivery carriers, they are considered as a new promising class of polymeric gene carriers with the following advantages.

  • A well-defined number of amine groups on the surface of the cationic dendrimers can coalesce nucleic acids into small nanoparticles through ionic interactions, protecting them from enzymatic degradation.
  • The high density of tertiary amine groups inside the dendrimer has a strong pH buffering capacity (pKa∼6.0), which facilitates efficient endosomal escape of dendrimer/DNA polyplexes from the endocytic pathway through the possible proton-sponge mechanism.

Dendrimer-Lipid Conjugation Improves Gene Transfection Efficiency

Gene transfection suffers from barriers such as multimer instability, insufficient cellular uptake and endosomal escape, difficulties in DNA unpacking, and DNA degradation by cytosolic nuclease. These barriers can be broken when cationic dendrimers are modified with various functional ligands (lipids, amino acids, peptides, and proteins, etc.). Lipid modifications are widely used to improve the performance of various polymeric gene vectors. When dendrimers are modified by lipids, efficient gene delivery can be achieved through direct binding of lipids to nucleic acids. Balancing the cationic charge and lipid content of a polymeric gene vector improves both cellular uptake and endosomal escape of the polyplexes. Lipid modification has a beneficial effect on dendrimer stability, serum stability and intracellular dissociation from DNA. In addition, dendrimer modification by lipids can lead to specific cellular uptake by endothelial cells via caveolae-mediated endocytosis. For example, when dendrimers are conjugated with fatty acids (e.g., lauric acid, myristic acid and palmitic acids), the cellular uptake of dendrimer/DNA multimers is increased due to the presence of lipids, showing greatly increased gene transfection efficacy on mesenchymal stem cells.
