Introduction into Cyclodextrins

As an oligosaccharide, cyclodextrins (abbreviated CDs) are produced from starch by an enzymatic reaction and are sugar molecules bound together in rings of different sizes, specifically these sugar units are known as glucopyranosides. They are used in food, pharmaceutical, drug delivery, chemical industry, agriculture, and environmental engineering. Cyclodextrins consist of six or more glucose units linked by α-1,4-glucosidic bonds. Typical cyclodextrins contain 6, 7 or 8 glucose units, named α-, β- and γ-cyclodextrin, respectively, and are generally regarded as safe. Among them, β- cyclodextrins are the most studied and most frequently used due to their low cost, availability, and their ability to encapsulate a wide range of molecules in their inner lumen. Cyclodextrins are often used as enhancers of gene and drug delivery, protect oligonucleotides from nucleases, and facilitate endosomal escape of pDNA complexes after cellular uptake. In addition, the variable reactivity of hydroxyl groups and remarkable encapsulation properties can improve the physical, chemical, and biological properties of host molecules, making cyclodextrins perfect candidates for the construction of drug delivery systems.
Dendrimer-Cyclodextrin Conjugation as Delivery Vectors
Nanomedicine has shown great potential for drug delivery and targeted release due to its ability to improve safety by reducing off-target organ delivery and toxicity to tissues. As a nanomaterial, dendrimers have many unique properties, such as monodispersity, many functional groups on the surface, and an excellent ability to encapsulate host molecules in a hydrophobic environment, making them ideal nanocarriers for targeted delivery of therapeutic and diagnostic drugs. Cyclodextrins have hydrophobic interiors and hydrophilic exteriors and can form complexes with hydrophilic compounds. When dendrimers are conjugated with cyclodextrins, cyclodextrins can confer higher solubility and stability to drugs, and their internal cavities can encapsulate various lipid molecules (cholesterol, bile acids, etc.) to improve the affinity to cell membranes. In addition, the solubility, stability, and biocompatibility of dendrimers can be improved. Therefore, dendrimers are excellent drug delivery and gene delivery carriers when conjugated with cyclodextrins.
